概要
Our research focuses on temporally dynamic mechanisms underlying T cell differentiation and responses. In the seminar, I will discuss that T-cell activities are controlled by temporally dynamic mechanisms, which can be revealed by single cell-level analysis. Firstly, I will discuss recent findings by single cell RNA- seq analysis, which shows that T-cells dynamically and gradually change into activated, effector, and regulatory phenotypes in cancer microenvironments (1) and in COVID-19 patients (2). Thus, I will demonstrate that immune dysregulationcanbeunderstoodasalteredT-cellactivationdynamics. Next,I will show our recent findings using Timer of Cellular Kinetics and Activity(Tocky), which analyses temporal changes in gene expression and thereby reveals temporal dynamics of T-cell activities in vivo (3, 4). I will show data from thymic development and cancer, highlighting the importance of temporal dynamics of T-cell receptor signalling downstream activities. Lastly, I will discuss the new model of T-cell regulation that temporally dynamic feedback mechanisms of activated T-cells self-regulate themselves and control activities of other T-cells.
